Abstract:
Im Rahmen dieses Promotionsprojektes wurde die Wirkung von Dexmedetomidin auf HCN-Kanäle in thalamokortikalen Schaltneuronen (TC-Neurone) untersucht, welche bereits als Zielstruktur für andere Anästhetika identifiziert worden sind. Dafür wurden in vitalen Hirnschnittpräparaten von Mäusen TC-Neurone mittels Patch-Clamp-Technik abgeleitet. Dexmedetomidin hatte in klinisch relevanten Konzentrationen weder einen Einfluss auf die Erregbarkeit der TC-Neurone noch auf die Leitfähigkeit der HCN-Kanäle. Erst bei höheren Konzentrationen zeigte sich eine Hemmung der HCN-Kanäle, welche sich jedoch am ehesten durch unspezifische Arzneimittelinteraktionen erklären lässt.

HCN-channels in thalamocortical relay neurons (TC neurons) were already identified as targets by different anesthetics. In this thesis, the effect of dexmedetomine on TC neurons was examined using the patch-clamp-technique in acute murine brain slices. When using clinically relevant concentrations, no effect could be shown on the excitation of TC neurons and the conductivity of HCN-channels. Only higher concentrations resulted in an inhibition of HCN-channels, which can most likely be explained by unspecific drug interactions.
