Abstract:
The aim of this work was to develop a novel MYC DEGrader (MDEG) based on the PROTAC technology. MDEG-541 was identified as the most potent compound and a treatment of cancer cells led to reduced cell viability. The dose- and time-dependent degradation of tumor-related proteins MYC, GSPT1, GSPT2 and PLK1 was dependent on the CRBN, the proteasome and ubiquitination. Further, the efficiency of MDEG-541 was tested in 3D organoid models. MDEG-541 offers a promising and novel strategy to target multiple cancers.

Die Substanz MDEG-541 zeigte in diesem Screen die höchste Effektivität und eine Behandlung von Krebszellen führte zur Reduktion der Zellviabilität. Die Tumor-assoziierten Proteine MYC, GSPT1, GSPT2 und PLK1 wurden abhängig von CRBN, dem Proteasom und der Ubiquitinierung inhibiert.
