Innate immunity contributes to allergic asthma with macrophages as orchestrators. In this thesis, we show that house dust mite shifts the macrophage eicosanoid profile from lipoxygenase- to cyclooxygenase-derived mediators. Repeated exposure to HDM leads to a trained immunity program of high CCL17 and leukotrienes, reflected in macrophages of mite-allergic patients. A helminth extract of Heligmosomoides polygyrus bakerii alleviates allergic airway inflammation via an antiinflammatory switch in macrophages characterized by high prostaglandin E2 and IL-10. Thus, macrophages are implicated in pathomechanisms of allergic asthma, but also a promising target for immunomodulation.
