Abstract:
Untersucht wurde die Immunogenität dezellularisierter Rattennieren als Bioscaffolds in der art- und gewebeüberschreitenden Tissue Engineering-Anwendung. Nach Perfusionsdezellularisierung mit SDS-Lösung zeigte sich eine deutlich reduzierte Immunogenität der Biomatrices für humane Immunzellen. Humane und Rattenosteoblasten konnten auf den Biomatrices proliferieren und behielten ihre Differenzierung bei. Die Arbeit untermauert, dass diese Dezellularisierungstechnik das Potential hat, eine Plattformtechnologie für das Tissue Engineering zu werden.

Aspects of biocompatibility of decellularized rat kidneys as biomatrices for cells from different tissue and species types were evaluated. There was a significant reduction in immunogenicity of the biomatrices for human immune cells after perfusion decellularization with SDS. Also, human and rat osteoblasts proliferated and kept their differentiation in culture with the biomatrices. These findings support our goal of developing this decellularization method into a platform technology even for cells of different tissue and species types.
