The high pressure induced stress response of bacteria was investigated for gram positive bacteria by Lactobacillus sanfranciscensis using 2-D gel electrophoresis. The stress response was compared to those of osmo, acid and heat stress and to cells of the stationary growth phase as well. Of 30 high pressure induced proteins six were identified as ClpL, GroEL, RbsK, GapDH, AdhE and a seryl-tRNA-synthetase. The high pressure induced stress response of gram negative bacteria was investigated by Escherichia coli using DNA micro array. Gram negative bacteria showed a similar stress reponse compared to those of grampositive bacteria. For the characterization of the high pressure induced stress reponse, the suitability of Green Fluorescent Protein (GFP) as a marker protein in fusion genes for the in situ measurement under high pressure was investigated. Purified protein was resistant to high pressure treatment to a pressure of 1.0 GPa. It was only possible to express GFP in Escerichia coli.
