Myenteric nervous tissue was isolated from the small intestine of adult rats and brought into primary cell culture for up to 14 days. Growth and differentiation under culture conditions was characterized. Neurons, nNOS-positive Neurons and glial cells were identified by immunochemical staining. NO-dependent modulation of calcium signaling in human colonic myocytes was studied by fluorimetric calcium imaging. A NO-induced, cGMP-dependent inhibition of inositoltriphosphate-dependent calcium release was demonstrated in human colonic myocytes.
