Abstract:
Multiples Myelom wird effektiv durch Inhibition des Ubiquitin Proteasom Systems behandelt. Als zugrundeliegender, krankheitsspezifischer Mechanismus wird hier erstmalig die Deregulation der mTOR Signalwege über die Ubiquitin-Ligase Fbxo9 und die Kinase CK2 identifiziert, wobei Fbxo9-Expression in Myelomzellen mit Überleben und besserem Ansprechen auf Proteasominhibitoren korreliert. Translated abstract:
Multiple Myeloma is effectively treated by inhibition of the ubiquitin proteasome system. The deregulation of mTOR signalling pathways via the ubiquitin ligase Fbxo9 and the kinase CK2 is here newly identified as the underlying, myeloma-specific mechanism, whereas Fbxo9 expression in myeloma cells correlated with survival and better response to proteasome inhibitors. As such, Fbxo9 and CK2 are characterised as novel therapeutic target structures as well as biomarker in Multiple Myeloma.
