Im Gastrointestinaltrakt bildet das intestinale Epithel die wichtigste Barriere zwischen Bakterien und Antigenen des Darmlumens einerseits und Immunzellen der Lamina propria andererseits. Diese Dissertation befasst sich mit der Rolle ausgewählter, auf intestinalen Epithelzellen exprimierter Chemokin- sowie Typ I- und Typ II-Zytokin-Rezeptoren. Im Einzelnen wurden ausführlich die Expression, Signaltransduktion und die spezifischen biologischen Funktionen dieser Rezeptor-Liganden-Systeme charakterisiert. Es zeigte sich, dass CXCL12/CXCR4 seine Hauptfunktionen in der kolorektalen Kanzerogenese sowie der Metastasierung von kolorektalen Tumorzellen aufweist. Die Chemokine CCL20 und CXCL16 sowie die Zytokine IL-31, IL-22 und IL-26 vermitteln intestinale Entzündung und sind bei Patienten mit chronisch entzündlichen Darmerkrankungen verstärkt exprimiert. IL-22 trägt außerdem zur Aufrechterhaltung und zum Schutz der intestinalen Barriere bei.     «

In the gastrointestinal tract, the intestinal epithelium forms an important barrier between bacteria and antigens of the intestinal lumen on the one hand and immune cells of the lamina propria on the other hand. In the present work, the role of selected chemokine receptors and type I and type II cytokine receptors that are expressed by intestinal epithelial cells was investigated. In detail, a comprehensive analysis of the expression, signal transduction and the specific biological functions of these receptor/ligand systems was performed. While CXCL12/CXCR4 play a major role in colorectal carcinogenesis and in metastasis of colorectal tumors, the chemokines CCL20 and CXCL16 as well as the cytokines IL-31, IL-22 and IL-26 mediate intestinal inflammation. Their expression is upregulated in patients with inflammatory bowel diseases. In addition, IL-22 contributes to the maintenance and protection of the intestinal barrier.     «
