Abstract:
In dieser Arbeit wurden für biomedizinische Anwendungen Anticalin-Proteine gegen die extrazellulären Domänen von humanem und murinem CD98hc aus einer kombinatorischen Genbibliothek selektiert und mittels Protein-Engineering bezüglich ihrer biophysikalischen Eigenschaften optimiert. Strukturelle Untersuchungen der CD98hc-Proteine aus Mensch und Maus sowie ihrer Komplexe mit den entsprechenden Anticalinen lieferten Einblicke in die biomolekulare Erkennung dieser Membranproteine. Die Eignung des human-spezifischen Anticalin-Leitkandidaten für die translationale Forschung wurde durch Experimente mit Krebszelllinien in vitro und im Mausmodell nachgewiesen.

In this work, Anticalin proteins for biomedical applications were selected towards the extracellular domains of human and murine CD98hc using a combinatorial gene library and optimized with regard to their biophysical properties through protein engineering. Structural studies of the murine and human CD98hc proteins as well as their complexes with the respective Anticalins provided insights into the biomolecular recognition of these membrane proteins. The potential of the human-specific Anticalin lead candidate for translational research was demonstrated by experiments with cancer cell lines in vitro and in mouse models.
