Kurzfassung:
Angelehnt an das Konzept bispezifischer Antikörper wurden bispezifische Enzyme zum Abbau von Organophosphat-Verbindungen (OPs) durch Ausnutzen der homodimeren Quartärstruktur von bakteriellen Phosphotriesterasen (PTE) generiert. Verschiedene Untereinheiten von PTE-Varianten aus Brevundimonas Diminuta sowie Agrobacterium radiobacter wurden erfolgreich kombiniert und deren Heterodimerisierung durch rationales Protein-Engineering stabilisiert. Der resultierende Bioscavenger aus PTEs unterschiedlichen bakteriellen Ursprungs weist ein beispiellos breites Substratprofil gegenüber OP-Kampfstoffen und -Pestiziden auf.

Inspired by the concept of bispecific antibodies, bispecific enzymes for the detoxification of organophosphates (OPs) were created by exploiting the homodimeric quarternary structure of bacterial phosphotriesterases (PTEs). Subunits of PTE variants from Brevundimonas diminuta and Agrobacterium radiobacter were successfully combined and their heterodimerization was stabilized by rational protein engineering. The resulting bioscavenger from PTEs with different bacterial origin exhibits an unprecedented broad OP substrate profile towards nerve agents and pesticides.
