Rapid identification and differentiation of strains from bacteria belonging to the same species become more and more important. However, physiological and biochemical methods for identification are often time-consuming and unreliable. Consequently molecular diagnostic methods, based on PCR-techniques and probe hybridization procedures play an important role in identification. It was possible to construct strain specific primer pairs for several human pathogenic legionellae, plant pathogenic Xanthomonas campestris and biotechnologically important streptomyces strains using a subtractive hybridization method in microwell plates (MASH). Based on the systems of MASH and reverse hybridization, a molecular screening technique was developed to search for specific genes and their variants. By using this newly developed method it was also possible to find and to isolate genes coding for virulence factors.
