Paenibacillus polymyxa is a Gram-positive soil bacterium that has been of biotechnological interest for the production of highly pure butanediol as well as exopolysaccharides (EPS). In this dissertation the butanediol pathway was metabolically engineered to increase product titers. Furthermore, the structure of the EPS was elucidated by a combinatorial CRISPR-Cas9 mediated knock-out approach and rheologically characterized. Finally, a novel Cas12a based tool for transcriptional activation and repression was developed.
