The aim of this study is to elicit the influence of high pressure treatment on the allergenicity of foods. Several birch pollen allergy related foods were selected for testing. The selection included hazelnut, carrot, cherry, peach, celery and apple. Native and high pressure treated samples (300 MPa and 600 MPa and 5 minutes dwell) were tested by means of prick to prick test in allergic patients. In general, a decrease of allergy symptoms due to high pressure treatment could be observed. An inactivation of the Bet v 1 homologue allergens may be responsible for this effect. Nevertheless, some of the minor allergens seem to be more high pressure resistant. Thus, some patients sensitized to these allergens develop allergic skin reaction despite high pressure treatment.To investigate mechanisms of high pressure allergen inactivation, apples and the major apple allergen Mal d 1 were analyzed in vitro and in vivo. Prick to prick test results were affirmed by oral provocation test. In a placebo controlled blinded provocation study 19 / 19 allergic patients, reporting on symptoms after apple consumption, tolerated high pressure treated apple (600 MPa, 5 minutes) without symptoms. Extensive prick to prick tests show a correlation between allergen inactivation and applied pressure altitude.Recombinant Mal d 1 was dissolved in neutral buffer and transmuted to native conformation. Native and high pressure treated aliquots were analyzed by means of spectroscopy. CD and FTIR spectra showed high pressure induced structural alterations.
