Abstract:
Der Lipid-Stoffwechsel ist eine wichtige Quelle flüchtiger Aromastoffe im Kulturapfel. Aroma-relevante Aldehyde, Alkohole und unverzweigte Ester entstehen in der Frucht vornehmlich durch den Lipoxygenase-(LOX)-Stoffwechselweg. In dieser Arbeit wurden verschiedene rekombinante LOX des Apfels biochemisch charakterisiert. Die chromatografische Analyse ihrer Produkte offenbarte Isoformen mit (R)-LOX-Aktivität. Durch zielgerichtete Mutagenese wurden die Determinanten dieser Spezifität bestimmt.
Translated abstract:
The lipid metabolism is an important source for aroma volatiles in cultivated apple. Lipoxygenases (LOXs) catalyse the primary reaction in the formation of aroma relevant aldehydes, alcohols and esters during fruit ripening. In this study, recombinant proteins of several apple LOXs were biochemically characterised. The analysis of their products revealed isoforms with (R)-LOX activity. Determinants of that specificity were determined via site-directed mutagenesis.
