- Abstract:
- While they are histologically homogeneous, the structural organization of the liver (zonation), natural genome duplications (polyploidization), and intracellular lipid accumulation (hepatic steatosis) are known sources of heterogeneity among hepatocytes. This thesis studies cellular heterogeneity through single cell transcriptome sequencing. The results show that polyploidy and zonation are intertwined in vivo and that hepatocytes retain cellular heterogeneity in vitro in absence of zonation.

- Obwohl sie histologisch homogen sind, stellen die Leberstruktur (Zonierung), natürliche Genomduplizierung (Polyploidie) und intra-zelluläre Fettanlagerung (Fettleber) bekannte Quellen von Heterogenität zwischen Hepatozyten dar. Diese Dissertation erforscht zelluläre Heterogenität mittels Sequenzierung des Transkriptoms einzelner Zellen. Die Ergebnisse zeigen, dass Polyploidie und Zonierung in vivo verbunden sind und Hepatozyen in vitro Heterogenität behalten in Abwesenheit von Zonierung.
