Abstract:
Das 20S Proteasom ist eine ca. 700 kDa schwere zylinderförmige, multikatalytische Protease, die von einem zentralen Kanal durchzogen ist. Dieser weitet sich zu drei internen Kammern auf, zwei Vorkammern und einer zentralen Kammer, in der sich die aktiven Zentren befinden. Zur Untersuchung des Mechanismus der Substrattranslokation in das 20S Proteasom wurden "Host-Guest" Komplexe des inhibierten 20S Proteasoms aus Thermoplasma acidophilum mit Cytochrom c und GFP hergestellt. The 20S proteasome is a 700 kDa cylinder shaped proteolytic complex. It is traversed by a central channel, which widens into three cavities, two antechambers and one central chamber which harbours the active sites. In order to elucidate the mechanism of substrate translocation into the 20S proteasome, "host-guest" complexes of the inhibited 20S proteasomes from thermoplasma acidophilum with cytochrome c and gfp were formed.
