This dissertation investigates the consequences of therapeutic B-cell depletion with Rituximab in multiple sclerosis and NMO. Monocytes from patients were isolated before and after the treatment and tested for their reactivity. Additionally, regulatory T-cell frequency was measured. Before and after therapy the variability of reactivity was significantly increased between the patients. Apparently, some patients in these indications may benefit from a Rituximab therapy while others may be at risk. Subsequently, an intrathecal application study in a mouse model was performed.
