Die vorliegende Arbeit zeigt am Beispiel eines in-vitro Co-Kultur Modells aus hämatopoetischen Stamm- und Progenitorzellen von MDS-Patienten (MDS-HSPZ) und Stromazellen, dass MDS-HSPZ die supportiven Eigenschaften der sie umgebenden Stromazellen zu ihren Gunsten beeinflussen können. Als ein Schlüsselprotein hierbei wurde Osteopontin identifiziert, dessen Expression durch MDS-HSPZ in Stromazellen induziert werden kann. Weiterhin wird dargestellt, dass Stromazellen, die mit dem epigenetisch wirksamen 5-Azacitidine behandelt wurden, den Support gegenüber MDS-HSPZ reduzieren, was sich in deren Genexpression, Zellzyklusregulation, Apoptose- und Differenzierungsverhalten widerspiegelt. Somit ist von einem bisher unbekannten Wirkmechanismus des Pharmakons auszugehen.
