Abstract:
In this thesis I show crystal structures of the FGFR4 kinase. Two apo-structures have been solved in different crystal forms, and two structures with inhibitors (Dovitinib/Ponatinib) that are currently used in clinical trials. The structures show an autoinhibited conformation of the apo-protein and the interactions of the inhibitors with the kinase. The second part covers the establishment of expression and purification protocols for the human ORAI1 calcium channel for structural studies.

In dieser Arbeit zeige ich Kristallstrukturen der humanen FGFR4 kinase, zwei Strukturen des apo-Proteins in unterschiedlichen Kristallformen, und zwei Strukturen im Komplex mit Inhibitoren, die zur Zeit in klinischen Studien getestet werden. Die Strukturen zeigen eine autoinhibiertes apo-Protein sowie die Interaktionen der Kinase mit den Inhibitoren. Im zweiten Teil beschreibe ich die Etablierung von Expressions- und Reinigungsprotokollen für die strukturelle Analyse des Kalziumkanals ORAI1.
