Experimentelle und erste klinische Studien haben gezeigt, dass die intrakoronare Gabe von autologen Stammzellen und endothelialen Progenitorzellen potentiell die Herzfunktion nach akutem Herzinfarkt verbessert. Inwieweit eine Expansion der endothelialen Progenitorzellen bei Patienten mit koronarer Herzerkrankung über Wochen hinweg möglich ist, wird in der Arbeit untersucht. Dazu wurden CD34+ Zellen von 65 Spendern in verschiednen Stadien der koronaren Herzerkrankung isoliert und in endothelialem Medium expandiert. Es zeigte sich, dass die Expansion prinzipiell möglich ist, es wurden aber krankheitsspezifische Unterschiede der Expansionsfähigkeit beobachtet. Weiter Untersuchungen zeigten eine Beeinträchtigung des Proliferationspotentials der endothelialen Progenitorzellen durch Einfriervorgänge.     «

Initial and experimental studies have shown, that the intracoronary application of autologous stem cells and endothelial progenitor cells have the potential to improve the heartfunction after acute myocardial infarction. To what extend an expansion of the endothelial progenitor cells with patients with coronary heart disease over weeks is possible, will be examined in this study. For this, the CD34+ cells from 65 donors in different stages of coronary illness were isolated and in an endothelial medium expanded. It was shown that the expansion was possible in principal. Diseasse-specific-differences of expansion possibility were observed. Further investigations showed limitations of the proliferation potential of the endothelial progenitor cells through freezing processes.      «
