Reversible switchable proteins (RSPs) provided a better alternative for optoacoustic (OA) imaging due to its signal modulation. In this work, a broad range of RSPs was characterized for their photophysical properties in OA. Later, two new superior rsOAPs were developed based on BphPs. A highly robust machine learning-based software for commercial imaging systems was used for data analysis. Using same, a highly sensitive, multiplexed visualization of T lymphocytes, bacteria, and tumors in the mouse body and brain was demonstrated.
