Epigenetics and approaches for targeted therapies provide unique opportunities for targeted treatment of a plethora of different diseases. Here, we analyzed the epigenetic landscape of ES by inhibition of BET bromodomain proteins (BRD`s) and CDK9. JQ1 effectively inhibited ES cell and tumor growth and activated apoptotic mechanisms by inhibition of BRD function. I-73 another small molecule blocking the activity of CDK9 consistently inhibited growth and activated apoptotic mechanism. In combination therapy set ups, JQ1 and I-73 effectively block proliferation and tumor growth at low dosages.
