Abstract:
Kallikrein-ähnliche Peptidase 4 beeinflusst die Progression des Prostatakarzinoms. Die Mechanismen sind weitestgehend ungeklärt. In dieser Arbeit wurde eine differentielle Expression von COL1A2 und SPARC in prä-pro-KLK4 exprimierenden PC-3-Zellen verglichen zu Vektorkontrollzellen nachgewiesen. Darüber hinaus wurden die beiden Gene auch in PC-3-Zellen mit furinaktivierbarer und inaktiver KLK4, sowie in An- bzw. Abwesenheit eines spezifischen KLK4-Inhibitors differentiell exprimiert. Diese Ergebnisse legen eine proteolyseunabhängige Genregulation von SPARC und COL1A2 durch KLK4 in PC-3-Zellen nahe.

The kallikrein-related peptidase 4 has been proposed to affect tumor progression in prostate cancer. The underlying mechanisms are largely unknown. We confirmed differential gene expression of COL1A2 and SPARC in PC-3 cells transfected with pre-pro-KLK4 compared to vector control cells. Differential expression of those two genes could also be shown in PC-3 cells expressing furin-activatable and inactive KLK4 as well as in PC-3 cells cultivated in presence or absence of a specific KLK4 inhibitor. This suggests a proteolysis-independent gene regulation of SPARC and COL1A2 by KLK4 in PC-3 cells.
