Abstract:
In this work, a novel fluorescence-based biosensor is used to resolve very fast and slow binding kinetics in three case studies. Fast kinetics of 15 congeneric ligands against cAMP-dependent protein kinase are determined with a competition assay. In a second study, the transition dynamics of mono- to multivalent TNFa and its interplay with therapeutic antibodies is investigated. In a third example, the unusually slow dissociation of Sirtuin-rearranging ligands from Sirtuin-2 is studied.
