Abstract:
Accumulating evidence suggests that the Amyloid β peptide (Aβ) initiates the actions that lead to the observed neuronal death in Alzheimer’s disease. While the precise mechanism is not known, it has been proposed that Aβ could directly damage cellular membranes.
In this study, a combination of fluorescence microscopy, Focussed Ion Beam milling and cryoelectron tomography suggested that toxic Aβ aggregates do not directly damage the plasma membrane or fragment the golgi apparatus. Interestingly, the aggregates seem to be internalized by macropinocytosis.

Zahlreiche Forschungs- und Studienergebnisse lassen vermuten, dass das Amyloid β Peptid (Aβ) für den beobachteten neuronalen Zelltod im Gehirn von Alzheimer Patienten verantwortlich ist. Der genaue Mechanismus ist weitestgehend unbekannt, vorgeschlagen wurde allerdings unter anderem, dass Aβ zelluläre Membranen direkt schädigen könnte.
Diese Arbeit verwendete eine Kombination aus Fluoreszenzmikroskopie, Ionenstrahldünnung and Kryoelektronentomographie zur Überprüfung dieser Hypothese. Die Ergebnisse deuten daraufhin, dass die zellschädigenden Aβ Aggregate weder die Zellmembran direkt schädigen noch zu einer Fragmentierung des Golgi Apparats führen. Stattdessen scheinen die Aggreagte interessanterweise von den Zellen mittels Makropinozytose aufgenommen zu werden.
