- Abstract:
- Colorectal cancer (CRC) is a major cause of cancer related deaths worldwide. Pigs genetically predisposed to CRC model early stages of the human disease but further mutations are required to recapitulate advanced disease. Using CRISPR/Cas9 multiplexing, tumour suppressors were inactivated in colonic organoids from these pigs. Additionally, we activated a silent KRAS-G12D allele. Transformed organoids exhibited hallmarks of cancer cells and will be used for autologous transplantation.

- Darmkrebs (DK) ist eine Haupttodesursache bei Krebserkrankungen weltweit. Schweine mit einer genetischen Prädisposition für DK modellieren frühe Stadien der Erkrankung, aber weitere Mutationen sind nötig, um Folgestadien abzubilden. Wir nutzten CRISPR/Cas9-Multiplexing, um Tumorsuppressorgene in Darmorganoiden dieser Schweine zu inaktiviert. Zusätzlich aktivierten wir ein latentes KRAS-G12D Allel. Transformierte Organoide ähneln Krebszellen und werden zur autologen Transplantation eingesetzt.
