HT29 Zellen bilden in Zellkultur "tight junctions" (TJ) nach Behandlung mit Endopeptidasen. Dieser Prozess erfolgt innerhalb 3-5 Minuten, erfordert Protease Aktivität und ist temperatur-abhängig, was auf die Interaktion zellulärer Proteine und Lipide hinweist. Alternativ entstehen TJ durch Behandlung von HT29 Zellen mit hypertonen chaotropen Salzen z. B. (NH4)2SO4. Translated abstract:
Formation of tight junctions (TJ) can be induced in cultured cells HT29 by treatment with endopeptidases. This process occurs within 3-5 minutes, requires protease activity and is temperature dependent, indicating interaction of cellular proteins and lipids. Alternatively TJ formation can be induced by hypertonic chaotropic salts e.g. (NH4)2SO4. This process is inhibited by serine protease inhibitors (e.g. PMSF) indicating, that cellular proteases are involved in TJ formation.
