Abstract:
In dieser Arbeit wurden neue Glykosidhydrolasen aus überwiegend thermophilen Organismen heterolog in E. coli exprimiert und mit verschiedenen Modellsubstraten auf Aktivität untersucht. Es wurden hitzestabile α-L-Rhamnosidasen aus GH106 charakterisiert, und die Funktion und erste Struktur von β-D-Galactofuranosidasen mit α-L-Arabinofuranosidaseaktivität aus GH159 untersucht. Translated abstract:
In this work, novel glycoside hydrolases from predominantly thermophilic organisms were heterologously expressed in E. coli and screened for activity using various model substrates. Heat-stable α-L-rhamnosidases from GH106 were characterized, and the function and initial structure of β-D-galactofuranosidases with α-L-arabinofuranosidase activity from GH159 were investigated. In addition, the different functionality of modules of the arabinoxylan arabinofuranohydrolase Ts_Axh43A was described.
