- Im Rahmen dieser Arbeit wurde die Totalsynthese des makrozyklischen Thiopeptidantibiotikums GE2270 A in 20 Syntheseschritten (längste lineare Sequenz) und mit einer Gesamtausbeute von 4.8% durchgeführt. Die von uns eingesetzte konvergente Synthesestrategie basiert auf regioselektiven, Palladium-katalysierten Kreuzkupplungen hochkomplexer Fragmente an 2,6-Dibrom-3-iodpyridin. - Translated abstract:
- The total synthesis of the macrocyclic thiopeptide antibiotic GE2270 A was achieved in 20 steps (longest linear sequence) with an overall yield of 4.8%. Our synthetic strategy relied on regioselective, Palladium catalyzed cross-coupling reactions of advanced intermediates with 2,6-dibromo-3-iodopyridine. It turned out that the formation of the 29 membered macrocycle by intramoleculare Stille reaction is much more efficient than the classical macrolactamization.
