Abstract:
Arbeiten im Rahmen der Dissertation ermöglichten die Aufklärung der räumlichen Struktur der 3,4-Dihydroxy-2-butanon-4-phosphat Synthase aus M. jannaschii. Durch kinetische Experimente mit mutierten Proteinen konnte einzelnen Aminosäuren des aktiven Zentrums Funktionen bei der Katalyse zugewiesen werden. Die molekularbiologischen und biochemischen Vorarbeiten ermöglichten die Lösung der dreidimensionalen Struktur des GTP Cyclohydrolase I Feedbackregulatorproteins (GFRP). The three-dimensional structure of 3,4-dihydroxy-2-butanone-4-phosphate synthase from M. jannaschii with its natural substrate ribulose-5-phosphate has been determined. By Kinetic characterisation of mutant proteins some amino acids located at the active site could be assigned to special functions in catalysis.
