Abstract:
The disease-related mitochondrial membrane proteins Bak and MPV17 are studied in lipid bilayers by NMR and other biophysical methods. For pro-apoptotic Bak, both its pore-forming and its resting state, as well as its interaction with the anti-apoptotic player BclxL are characterized in nanodiscs and liposomes. For MPV17, NMR studies on the monomer are conducted in micelles, while the presumably pore-forming oligomer is studied in a lipid bilayer revealing an important role for cysteines.

Die mitochondrialen Membranproteine Bak und MPV17 werden u.a. mittels NMR in einer Membranumgebung studiert. Für pro-apoptotisches Bak wird sowohl der inaktive als auch der aktive Poren-bildende Zustand und die Interaktion mit dem anti-apoptotischen BclxL in Nanodiscs und Liposomen untersucht. Monomeres MPV17 wird mittels NMR in Mizellen und das vermutlich Poren-bildende Oligomer in einer Lipid-Doppenschicht untersucht. Dabei konnte den Cysteinen eine wichtige Rolle zugeordnet werden.
