abstract = {Adenocarincoma of the lung (NSCLC) represents one of the most common cancer entities often exhibiting activating mutations of KRAS. We tested the requirement for anti-apoptotic MCL-1 for the survival of KRAS-driven NSCLC. In MCL-1 expressing cell lines, MCL-1 protects against apoptosis irrespective of any other BCL-2 protein. Using lox-stop-lox-KrasG12D Mcl-1floxed mice, we find a significant reduction in tumor burden upon MCL-1 deletion in cancer lesions. Individual cancer lesions escape this mechanism and exhibit elevated STAT3 activation suggesting compensatory JAK-STAT signaling. },
