The highly conserved AAA ATPase Cdc48 from budding yeast constitutes an ubiquitin-specific segregase, whose specificity in various cellular processes is determined by different cofactors. In this work it was shown that Cdc48 and its cofactor Ubx5 bind to the cullins Cdc53 and Rtt101, which assemble two different ubiquitin ligase complexes. Ubx5 mediates the binding of Cdc48 to Cdc53. In addition, Ubx5 recruits Cdc48 to Mms22, a substrate of Rtt101, which is essential for the progression through stalled replication forks.
