An important step in transferring an apoptotic signal into cells is the oligomerisation (cross linking) of proteins. The aim of this study was to experimentally induce oligomerisation of artificial fusion proteins (Gyr1x_FADD_Flag, DR5-DD_Gyr1_Flag, DR5-DD_Gyr3x_Flag), thereby allowing the ligand-independent activation of the apoptotic pathway in human cell lines. For the Fusion-Protein DR5-DD_Gyr1x_Flag, crosslinking could be demonstrated by using various amounts of Coumermycin. However, the expected induction of the apoptotic signalling cascade, ultimately resulting in cell death, could not be shown.
