The turnover of cyclin-dependent kinase inhibitors (CKI) is of major significance for hematopoietic stem cell (HSC) homeostasis. This study shows, that cyclin-dependent kinase subunit 1 (Cks1) as a part of the E3 ubiquitin ligase complex SCFSkp2 and as an essential suppressor of the protein levels of CKI, controls cycling, quiescence and self-renewal in HSC, probably through regulation of CKI from the CIP/KIP family. Additionally it was shown, that Cks1 is up-regulated by the constitutively active tyrosine kinase BCR-ABL in chronic myeloid leukemia and suppressed upon imatinib therapy.
