Die vorliegende Arbeit untersucht die Effekte der Zytostatika Topotecan, Etoposid, Docetaxel und Estramustinphopat auf die Hemmung der Zellproliferation und die Induktion von Apoptose bei den Prostatakarzinomzelllinien LNCaP, DU-145 und PC-3. Die Ergebnisse der Monotherapien wurden hinsichtlich der Proliferationshemmung durch Isobologramme mit den Kombinationstherapien Topotecan und Etoposid, Topotecan und Docetaxel, Topotecan und Estramustinphosphat verglichen. Die Induktion der Apoptose wurde bei der Kombination Topotecan und Docetaxel untersucht und mit den jeweiligen Monotherapien von Topotecan und Docetaxel verglichen. Bei der Proliferation zeigte vor allem die Kombinationstherapie Topotecan/Etoposid und Topotecan/Docetaxel additive und synergistische Wirkbeziehungen. Eine erhöhte Apoptoseinduktion erreicht die Monotherapie Toptocan und Docetaxel, jedoch nicht die Kombination dieser Chemotherapeutika.     «

In the present study the cytotoxic effect of the cytostatic drugs Topotecan, Etoposid, Estramustinphosphat and Docetaxel was tested on proliferation and apoptosis of three established human prostatic cancer cell lines LNCaP, DU-145 and PC-3. The antiproliferative results of the mono-therapies was compared to the combination of Topotecan and Etoposid, Topotecan and Docetaxel, Topotecan and Estramustinphophat using the isobologram method. The apoptotic effect was analyzed on the combination of Topotecan and Docetaxel and compared to the mono-therapies. The proliferation of Topotecan/Etoposid and Topotecan/Docetaxel schowed additivie and synergistic activity.
