ELISAs (enzyme-linked immunosorbent assay) can be performed on antibody microarray platforms. By screening multiple antibody-antigen interactions in parallel on a single substrate, the simultaneous detection of a multiplicity of analytes in one sample in a single experiment becomes possible. For the parallel quantification of microorganisms in water samples, array-based sandwich ELISAs were applied, which were performed at an automated chemiluminescence microarray readout system. The first part of the thesis deals with the development of a simple and reproducible surface chemistry for the covalent immobilization of antibodies to glass substrates. The second part focuses on the chioce of suitable specific antibodies and on the characterization of the measuring system.
