Die Arbeit beschäftigt sich mit der Entwicklung neuartiger Methoden zur Analytik von Mykotoxinen, speziell der äußerst kanzerogenen Aflatoxine, in Lebensmitteln. Die Synthese von isotopenmarkierten Aflatoxinen ermöglichte die Entwicklung eines gut validierten Stabilisotopenverdünnungs-Assays mittels Flüssigkeitschromatographie und Tandem-Massenspektrometrie. Der Hauptteil der Arbeit umfasst jedoch die Generierung Aflatoxin-spezifischer monoklonaler Antikörper (Immunogensynthese, Immunisierungen, optimiertes Hybridomzell-Screening, Antikörper-Charakterisierung). Anti-Aflatoxin-Antikörper mit verschiedenen Charakteristika und Bindungskonstanten von bis zu 10^11 L/mol konnten gewonnen werden. Weiterhin wurden regenerierbare Aflatoxin-Mikroarray-Chips hergestellt. Diese Technologie erlaubt die Entwicklung automatisierter immunanalytischer Techniken für die schnelle Mykotoxin-Analytik.      «

This work describes the development of novel methods for the determination of mycotoxins, especially the very carcinogenic aflatoxins, in foods. The synthesis of isotope-labeled aflatoxins enabled the development of a stable-isotope dilution assay using liquid chromatography and tandem-mass spectrometry. The main section of the work deals with the generation of aflatoxin-specific monoclonal antibodies including syntheses of immunogens, immunisations, optimised hybridoma cell screenings and antibody characterisation. Anti-aflatoxin antibodies with various characteristics and binding constants of up to 10^11 L/mol could be obtained. Furthermore, regenerable aflatoxin microarray chips could be generated. This technology enables the development of automated immunoanalytical techniques for the rapid analysis of mycotoxins in foods.      «
