Abstract:
Mutations in the Pink1 gene cause Parkinson`s disease. This thesis concentrated on the role of Pink1 in mitochondrial function of astrocytes. Respiratory analysis of primary astrocytes from Pink1 deficient mice revealed strong deficits. Moreover, a new Pink1 mouse model was generated during this thesis. Behavioral phenotyping of these mice, that carry the Q126P mutation in the Pink1 gene, as well as the respiratory profile of their astrocytes showed differences compared to the knock-out model. In conclusion, the obtained results indicate cell type as well as mutation specific effects of Pink1.

Mutationen im Pink1 Gen verursachen Morbus Parkinson. In dieser Arbeit wurde die Rolle von Pink1 in der mitochondrialen Funktion von Astrozyten untersucht. Die respiratorische Analyse primärer Astrozyten von Pink1 defizienten Mäusen zeigte klare Defizite auf. Im Verlauf der Arbeit wurde zudem ein neues Pink1 Mausmodell generiert. Das Verhalten dieser Mäuse, die die Q126P Mutation im Pink1 Gen tragen, sowie die respiratorische Analyse ihrer Astrozyten, deuteten auf klare Differenzen zum Knock-out Modell hin. Somit weisen die vorliegenden Ergebnisse sowohl auf zelltyp- als auch mutationsspezifische Auswirkungen von Pink1 hin.
