Abstract:
In this thesis, commercially available basal lamina hydrogels are analyzed to study the effect of the biochemical composition of these gels on their biophysical properties. In addition, a thermoresponsive methylcellulose/mucin hybrid wound gel is developed where the methylcellulose compensates for the mechanical limitation of the anti-microbial mucins. Finally, to achieve orchestrated drug release from a hydrogel two molecular mechanisms are combined: i.e. a build-up of osmotic pressure by the depletion of a control molecule and triggered disaggregation of nanoparticle clusters by synthetic DNA sequences.

In dieser Arbeit wird der Effekt der biochemischen Zusammensetzung von kommerziell erhältlichen Basallamina-Hydrogelen auf ihre biophysikalischen Eigenschaften untersucht. Zudem wird ein thermoresposives Methylzellulose/Mucin Hybridwundgel entwickelt, bei dem die Methylzellulose die unzureichenden mechanischen Eigenschaften des antimikrobiellen Mucins kompensiert. Im letzten Teil werden zwei molekulare Mechanismen kombiniert um abgestimmte Medikamentenfreisetzung aus einem Hydrogel zu erreichen, zum einen Aufbau osmotischen Drucks durch die Verarmung eines Kontrollmoleküls zum anderen die initiierte Disaggregation von Nanopartikel-Clustern durch synthetische DNA-Sequenzen.
