Abstract:
Bisphosphonate hemmen den Stoffwechsel der Osteoklasten. Es wurde eine neue Methode entwickelt, um die tartratresistente, saure Phosphatase 5b (TRAP 5b), als Marker der Osteoklastenaktivität in humanen Knochen zu lokalisieren und zu quantifizieren. Patienten (n=12) der Kontroll- und Bisphosphonatgruppe (10mg Alendronat/d, 50d prä-OP) wurde der Hüftkopf durch eine Totalendoprothese ersetzt. Translated abstract:
Bisphosphonates inhibit osteoclast activity and induce apoptosis. A method was developed to quantify and localize tartratresistent acid phosphatase as a marker of osteoclast activity. Patients (n=12) of the control- and the bisphosphonate group (10mg alendronate/d, 50d prae-OP) underwent a total joint replacement of the hip. Samples of the femoral head area (A1), the femoral neck area- (A2) and the intertrochanteric area (A3) were harvested, grinded and TRAP 5b was analysed with an ELISA or were embedded in MMA, stained and the TRAP 5b-positive cells were counted. Bisphosphonate treatment leads to a moderate decrease in enzyme activity compared to the controls: 18% in area A3 (p<0.05), 5% in A2 and 11% in A1. Bisphosphonates may be incorporated and effective even in degenerative bone tissue. They might be able to inhibit osteolytic and degenerative processes in arthritic bone.
