In this dissertation the isopentenyl diphosphat precursor (E)-1-hydroxy-2-methyl-but-2-enyl 4-diphosphate was synthesized chemically. The identity of (E)-1-hydroxy-2-methyl-but-2-enyl 4-diphosphate with the product of the IspG protein was proved. Furthermore, the biological activity of (E)-1-hydroxy-2-methyl-but-2-enyl 4-diphosphate and structural analogues was studied using immunological methods. Hereby (E)-1-hydroxy-2-methyl-but-2-enyl 4-diphosphate was found to be an activator of the proliferation of Vgamma9/Vdelta2 T cells with an EC50 value of 70 pM. In cell cultures with (E)-1-hydroxy-2-methyl-but-2-enyl 4-diphosphate TNF-alpha and INF-gamma were produced.
