The TNF superfamily member LIGHT, described as a ligand for the LT beta receptor, HVEM, and DcR3, was inactivated in the mouse germline. LIGHT was shown to cooperate with LT beta in the organogenesis of the mesenteric lymph nodes but not in the generation of peripheral lymph nodes or a correct splenic microarchitecture. LIGHT acts synergistically together with CD28 in the rejection of allogeneic skin grafts in vivo. The underlying mechanism was defined in in vitro experiments showing reduced cytotoxic T cell responses, cytokine production, and CD8:CD4 ratios of activated T lymphocytes in the absence of LIGHT. Additionally, proliferative responses of LIGHT deficient splenocytes were decreased after T cell receptor stimulation. Thus, for LIGHT a cooperative role in lymphoid organogenesis and important costimulatory functions in T cell activation could be defined.
