Abstract:: Bei der heterologen Proteinproduktion im Organismus E. coli treten häufig Limitationen oder Wechselwirkungen mit der Zelle auf. Um diese Interaktion zu verstehen, wurde ein ganzheitlicher Ansatz verfolgt, der unter Belastung neben dem zellulären Metabolismus unterschiedliche Prozessstrategien und die zelleigenen Ressourcen betrachtet. Weiterhin konnte sowohl ein System zur Erfassung der zellulären Kapazität als auch ein modulares Plasmid, welches Expressionsdynamik visualisiert etabliert werden.
Translated abstract:: Limitations or interactions with the cell often occur during heterologous protein production in the host organism E. coli. In order to understand this interaction, a holistic approach was pursued, which, in addition to cellular metabolism under load, considers different process strategies and the cell's own resources. Furthermore, both a system for detecting the cellular capacity and a modular plasmid to visualize expression-dynamics could be established.
