Abstract:
This study involves the biochemical characterization of enzymes involved in the deoxyxylulose phosphate pathway of isoprenoid biosynthesis. The recombinant IspC proteins from A. thaliana and P. falciparum, IspE protein from A. aeolicus as well as IspF from A. thaliana were enzymatically characterized. The inhibition by chemical compounds was studied in comparison to the respective E. coli proteins. A mechanistic study of IspC proteins was carried out on the bases of NMR analyses.

Gegenstand dieser Arbeit ist die biochemische characterisierung von Enzymen des Desoxyxylulosephosphat-Wegs zur Isoprenoid-Biosynthese. Die rekombinanten IspC-Proteine aus A. thaliana und P. falciparum, das IspE-Protein aus A. aeolicus so wie IspF aus A. thaliana wurden enzymatisch charakterisiert. Die Hemmung durch chemische Verbindungen wurde im Vergleich zum jeweiligen Protein aus E. coli untersucht. Außerdem wurden IspC-proteine mechanistisch untersucht.
