Abstract:
Im Rahmen dieser Dissertation wurden zwei Aspekte zur Faltung und Assemblierung oligomerer Proteine bearbeitet. Zum einen wurden die Expression und die Sekretion des mono-klonalen Antikörpers MAK33 in der Hefe Saccharomyces cerevisiae analysiert. Die in das Kulturmedium abge-gebe-nen Antikörperketten (wenige µg/l) waren z.T. fragmentiert und formten keine aktiven Antikörper. Two aspects of folding and assembly of oligomeric proteins were investigated in this thesis. First, expression and secretion of the monoclonal antibody MAK33 in the yeast Saccharo-myces cerevisiae were analyzed. Small amounts of antibody chains (in the range of µg/l) were secreted into the culture medium. These chains were partially fragmented and did not assemble to active antibodies.
