In children with a first clinical event of inflammatory demyelinating CNS disease (acute disseminated encephalomyelitis (ADEM), clinically isolated syndrome (CIS)) as well as healthy and neurological controls, the occurrence of serum antibodies to the native CNS proteins myelin oligodendrocyte glycoprotein (MOG), proteolipid protein (PLP) and aquaporin-4 (AQP4) as well as the herpesviruses EBV, HHV-6 and HCMV was investigated. In a subgroup of children with ADEM and CIS, a specific IgG antibody response towards native MOG was detected. Moreover, these antibodies induced in vitro cytotoxicity of MOG expressing cells. IgM antibodies against MOG were only detected in some ADEM patients. Children with CIS showed elevated IgG antibody titers against EBV and HCMV which were not related to the antibody response against MOG.
