The clinically relevant probiotic mixture VSL#3 has been shown to be protective in the context of inflammatory bowel diseases (IBD) but the underlying protective mechanisms are unclear. The intestinal epithelium plays a major role in the maintenance of the intestinal homeostasis. Hence, the aim of the present thesis was to characterize structure-related anti-inflammatory mechanisms of probiotic bacteria in intestinal epithelial cells (IEC). It was found that a protease of Lactobacillus casei, lactocepin, mediates selective inhibition of the secretion of the chemokine IP-10 in IEC. This anti-inflammatory mechanisms was found to be physiologically relevant in two experimental mouse models for IBD.      «

Die probiotische Mischung VSL#3 ist im Bereich chronisch entzündlicher Darmerkrankungen (CED) klinisch relevant, die protektiven Mechanismen sind allerdings unbekannt. Das Darmepithel spielt bei der Aufrechterhaltung des Darmgleichgewichtes eine wichtige Rolle. Das Ziel der Doktorarbeit war somit die Charakterisierung strukturabhängiger anti-entzündlicher Mechanismen von VSL#3 in intestinalen Epithelzellen (IEC). Es wurde gezeigt, dass eine Protease aus Lactobacillus casei, Lactocepin, eine selektive Hemmung der Sekretion des Chemokins IP-10 aus IEC induziert. Dieser Effekt wurde in Mausmodellen für CED als physiologisch relevanter protektiver Mechanismus von VSL#3 identifiziert.      «
