Der Urokinase-Typ Plasminogen Aktivator Rezeptor (uPAR) ist für maligne Tumore wichtig. Ziel war es ein Ribozym zu entwickeln, das in der Lage ist, die mRNA von uPAR in vitro und in vivo katalytisch zu spalten und so in vivo die Expression des uPAR zu unterdrücken. Es sollte ein neues Verfahren entwickelt werden, um eine Library von Ribozymen in Bakterien auf ein solches Ribozym zu screenen. In dieser Arbeit wurde erstmals gezeigt, dass das Ribozym RZ-957 in vitro und in vivo die mRNA von uPAR schneidet und zu einer signifikanten Reduktion der Expression in Zellkultur führt. Dieser Effekt war in Bakterien nicht reproduzierbar.     «

The Urokinase plasminogen activator receptor (uPAR) plays an important role in cancer. The aim of this work was to develop a ribozyme which is able to cleave the mRNA of uPAR in vivo and in vitro to suppress the expression of uPAR in vivo. It was planned to develop a new method to screen a library of ribozymes in bacteria for such a ribozyme. This work shows for the first time that Ribozyme RZ-957 cleaves the mRNA of uPAR in vitro and in vivo and causes significant reduction of its expression in cell cultre. This effect was not reproduceable in bacteria.     «
