- Im Rahmen der vorliegenden Arbeit wurden Wege zur Totalsynthese von Geldanamycin untersucht. Trotz dessen Potenzial als Antitumor-Antibiotikum wurde bisher kein effizienter und flexibler Syntheseweg, der auch eine zielgerichtete Derivatisierung erlaubt, zu diesem Naturstoff realisiert. Im Zuge dieser Arbeit wurden stereoselektive Synthesestrategien für den Aufbau der stereogenen Zentren entwickelt und besonderer Wert auf neue Ringschlussvarianten zu der makrozyklischen Struktur des Geldanamycins... »
- Translated abstract:
- The following thesis describes new synthetic approaches to the ansamycin geldanamycin. Despite its potential as antitumor antibiotic there is still no efficient and flexible synthetic route to this natural product available, which also would make derivatives easily accessible. The conducted studies focused on synthetic strategies to build up the stereogenic centers and new methods to close the macrocyclic structure of geldanamycin.
